Seeing as it is Kenya's second largest city and premier port, Mombasa is pretty well-connected in terms of transport. You can fly here direct from Nairobi several times a day which land at Moi International airport and then transfer by ferry to South Coast which would take another 2 hours depending on the traffic and the wait at the ferry terminal.
Alternatively fly direct from Wilson Airport Nairobi direct to Ukunda Airstrip, a flight of about 1 hour and then about 20-30min to most of the South Coast Hotels.
The road between Nairobi and Mombasa is well-tarred and the ride should take about 10 hours. There is also a railway line running from Mombasa westwards - book a first-class ticket and hop aboard for an overnight train ride.
